ADX71943

ADX71943 is a selective positive allosteric modulator of gamma-aminobutyric acid subtype B (GABAB) receptors. A marketed product called baclofen works via GABAB activation and is approved to treat spasticity and is used off-label to treat a variety of other indications. Baclofen is a classsical orthosteric agonist and, although it has significant potential in a variety of other indications, its use has been limited because of the development of tolerance and dose-limiting side effects.

ADX71943 works via an allosteric mechanism and may avoid the development of tolerance and thus also avoid dose limiting side effects. As a result, this selective GABAB PAM has potential for a variety of indications including osteoarthritis, urinary incontinence and GERD.

ADX71943 is in preclinical testing and currently is scheduled to start Phase I testing in the fourth quarter of 2010.
